Yes, trains do run between Kyiv and Mykolayiv (Nikolaev is russian spelling) in winter. These are two major cities and administrative centers so there is good connection between them. You could try calling the Grand Tour travel agency in Lviv - I'm sure they would know more details. they also speak English: +380 322 769170
As far as booking from the USA - not sure if it's possible, but I suggest you buy a ticket once you are in Ukraine. This will be: 1. Cheaper 2. YOu sill save yourself an effort confirming this ticket in Ukraine
Tickets are easy to buy and especially in winter when trains are less crowded than in summer.
